The Gallery is located in the old city centre in Via Alloro, the most important street in the ancient Kalsa quarter, which became highly urbanized at the end of the 15th century because of its proximity to the harbour.

The museum is housed in a building designed by Carnalivari dating from the end of the 15th century, when it was the residence of Francesco Abatellis, Harbourmaster of the Kingdom. The palace is one of the most significant examples of Gothic-Catalan architecture in western Sicily. The palace was later converted into a convent and subsequently modified at various other times. Having been seriously damaged by bombing in 1943, it was restored by the Soprintendenza of Palermo and refurbished as a museum by Carlo Scarpa. It became the National (now Regional) Gallery in 1953-54.

The Gallery illustrates how art and culture developed in Palermo and western Sicily from the 12th to the 18th centuries.

The collections originate from purchases and donations and the acquisition of items from deconsecrated and suppressed convents which first belonged to the University Museum and then, after 1866, to the Palermo National Museum.
